The multi-dimensional roles of astrocytes in ALS
Despite significant progress in understanding the molecular and genetic aspects of amyotrophic lateral sclerosis (ALS), a fatal neurodegenerative disease characterized by the progressive loss of motor neurons, the precise and comprehensive pathomechanisms remain largely unknown. In addition to motor neuron involvement, recent studies using cellular and animal models of ALS indicate that there i...
متن کاملThe potential of GPNMB as novel neuroprotective factor in amyotrophic lateral sclerosis
Amyotrophic lateral sclerosis (ALS) is an incurable and fatal neurodegenerative disease characterized by the loss of motor neurons. Despite substantial research, the causes of ALS remain unclear. Glycoprotein nonmetastatic melanoma protein B (GPNMB) was identified as an ALS-related factor using DNA microarray analysis with mutant superoxide dismutase (SOD1(G93A)) mice. GPNMB was greatly induced...
متن کاملMotor Neuron Death in ALS: Programmed by Astrocytes?
Motor neurons in ALS die via cell-autonomous and non-cell-autonomous mechanisms. Using adult human astrocytes and motor neurons, Re et al. (2014), in this issue of Neuron, discover that familial and sporadic ALS-derived human adult astrocytes secrete neurotoxic factors that selectively kill motor neurons through necroptosis, suggesting a new therapeutic avenue.
متن کاملThe Role of Astrocytes in the Pathogenesis of Amyotrophic Lateral Sclerosis
Astrocytes are glial cells that play a crucial role in providing a supportive environment for neurons, mainly by cellcell interactions mediating numerous physiological and biochemical functions in the central nervous system (CNS). Astrocytes are also critical for intercellular signaling in the neurovascular unit. Although numerous reports have detailed the complex effects of astrocytes upon neu...
متن کاملNon-cell-autonomous effect of human SOD1 G37R astrocytes on motor neurons derived from human embryonic stem cells.
Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease characterized by motor neuron death. ALS can be induced by mutations in the superoxide dismutase 1 gene (SOD1). Evidence for the non-cell-autonomous nature of ALS emerged from the observation that wild-type glial cells extended the survival of SOD1 mutant motor neurons in chimeric mice. To uncover the contribution of astrocytes ...
متن کاملPotential link between aberrant astrocytes and paralysis in ALS
that dysfunctional cells in the nervous system known as astrocytes induce the motor neuron damage that characterizes amyotrophic lateral sclerosis (ALS). But researchers have been unable to identify an astrocytic phenotype that becomes toxic to motor neurons. Pablo DíazAmarilla et al. (pp. 18126–18131) isolated a previously unknown type of astrocyte with aberrant phenotypic features from primar...
